Growth hormone-releasing hormone (GHRH) analogues are synthetic peptides modelled on the endogenous 44-amino-acid hypothalamic releasing hormone, or on its shorter biologically active fragment. Compounds in this class are structural variants: sequences are truncated, substituted at positions prone to enzymatic cleavage, or modified at the terminus to alter stability in solution. Tesamorelin and the CJC-1295 series are the analogues most often encountered in the published literature.
Mechanistically, this family is studied for its interaction with the GHRH receptor, a class B G-protein-coupled receptor. Laboratory investigation typically centres on receptor binding affinity, downstream cyclic AMP signalling in transfected cell lines, and the comparative enzymatic stability of modified sequences against the unmodified parent peptide. Because the substitutions in this class are largely aimed at resisting dipeptidyl peptidase cleavage, degradation kinetics is a recurring theme in the preclinical record.
